This document describes a method for the determination of individual ergot alkaloids and tropane alkaloids in unprocessed cereals and cereal-based compound feeds by high performance liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). The ergot alkaloids covered in this method are: ergocornine, ergocorninine, ergocristine, ergocristinine, a-ergocryptine, a-ergocryptinine, ergometrine, ergometrinine, ergosine, ergosinine, ergotamine and ergotaminine. The tropane alkaloids covered in this method are: atropine (hyoscyamine) and scopolamine. The limit of quantification for all compounds should be at least 10 µg/kg. This method has been in-house validated in the range 10 - 500 µg/kg for individual alkaloids. The detection of concentrations above 500 µg/kg is possible via dilution of the sample extract, but this has not been validated.
